Human VPS34 (vacuolar protein sorting 34), GenBank Accession No. NM_002647, a.a. 2-888(end) with N-terminal FLAG-tag, expressed in Sf9 insect cells via a baculovirus expression system. MW=98.2 kDa. Specific Activity: 140 pmole/min/µg. Enzyme reaction was conducted in a buffer (40 µl) containing 40 mM Tris (pH 7.4), 10 mM MgCl2, 2 mM MnCl2, 0.1 mg/ml BSA, 100 µM ATP, 100 µM PI:PS substrate (BPS # BPS-40560) and 0.6-5 ng/µl Vps34 at 30°C for 20 min. Kinase reaction was measured using ADP-Glo reagents (Promega). Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
