Troponin, working in conjunction with tropomyosin, functions as a molecular switch, regulating muscle contraction in response to changes in the intracellular Ca2+ concentration. Troponin consists of three subunits, the Ca2+-binding subunit troponin C (TnC), the tropomyosin-binding subunit troponin T (TnT), and the inhibitory subunit troponin I (TnI). Troponin I binds to actin in thin myofilaments to hold the actin-tropomyosin complex in place, because of which myosin cannot bind to actin in relaxed muscle. When Calcium binds to the Troponin C it causes certain confomational changes which leads to dislocation of troponin I. The letter "I" is given due to its inhibitory character. Recombinant Human cTnI is a single, non-glycosylated, polypeptide chain containing 165 amino acids and having a molecular mass of 24,016D. , Tissue-specific Troponin Subtypes:, Slow-twitch skeletal muscle isoform troponin I, TNNI1 (1q31.3, 191042), Fast-twitch skeletal muscle isoform troponin I, TNNI2 (11p15.5, 191043), Cardiac troponin I, TNNI3 (19q13.4, 191044), Dimers and Aggregates: < 1% as determined by silver-stained SDS-PAGE gel analysis. Protein Content: Protein quantitation was carried out by:, 0.25-5.0mg/ml Bradford assay vs. BSA. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, add 0.1% HSA or BSA, aliquot and store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
