TLR4 Inhibitory Peptide Set (VIPER)
Bemerkungen: Set contains 1 mg each of VIPER and CP7 (control peptide). VIPER (viral inhibitory peptide of TLR4) is a TLR4 specific inhibitory peptide that potentently inhibits TLR4-mediated responses and is inert towards other TLR pathways (Bowie et al. 2010, Lysakova-Devine et al. 2010). VIPER consists of an 11 amino acid (KYSFKLILAEY)inhibitory sequence, derived from the A46 vaccinia virus protein, linked to a 9R (RRRRRRRRR) homopolymer delivery sequence. VIPER interferes with the interaction between TLR4 and its adaptor proteins Mal and TRAM. It is thought that VIPER inhibits TLR4 signaling by binding to the TIR domains of Mal and TRAM. Since TIR domains engage the MAl and TRAM adaptor proteins with TLR4, blocking adaptor TIR domains with VIPER would disrupt propagation of the TLR4 signal. However, the exact mechanism by which VIPER inhibits TLR4 signaling remains to be fully elucidated. Likewise, the specific VIPER binding sites on TRAM and Mal TIR domains remain to be identified.
