IHC, ISH Ready-to-use pepsin solution in stabilizing buffer. Pepsin is a commonly used digestive enzyme for immunohistochemical (IHC) and in situ-hybridization (ISH) procedures. Most of the commonly used fixatives cause cross-linking of proteins, which leads to masking or denaturation of tissue antigens. Treatment of fixed tissue sections with proteolytic enzymes as e.g. pepsin is often required to unmask antigens for optimal staining in IHC procedures. For ISH applications treatment with proteolytic enzyms is required to digest excess aldehyde linkages to enable DNA or RNA probes to penetrate the tissue sections.
