Interleukin-19 (IL-19) is a novel cytokine that was initially identified during a sequence data base search aimed at finding potential Interleukin-10 homologs. Interleukin-19 shares 21% amino acid identity with IL-10. The exon/intron structure of IL-19 is similar to that of the human IL-10 gene, comprising five exons and four introns within the coding region of the IL-19 cDNA. IL-19 shares a receptor complex with IL-20, indicating that the biological activities of these two cytokines overlap and that both may play an important role in regulating development and proper functioning of the skin. Unlike IL-10, which forms an intercalated dimer, the molecule of IL-19 is a monomer made of seven amphipathic helices, A-G, creating a unique helical bundle. Description: Recombinant Mouse IL-19 produced in E.Coli is a single, non-glycosylated polypeptide chain containing 152 amino acids and having a molecular mass of 17722 Dalton. Recombinant Human IL-19 is purified by proprietary chromatographic techniques. Amino Acid Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Met-Leu-Arg-Arg-Cys. Dimers and Aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis. Biological Activity: Recombinant Mouse IL-19 is fully biologically active when compared to standard. The biological activity was determined by measuring the dose-dependent activation of STAT3 in human epidermal keratinocytes (HEK2a cells). Significant STAT3 activation is observed with >100ng/mL of recombinant mouse IL-19. Endotoxin: < 0.1ng/ug (IEU/ug) of Mouse IL-19. Protein Content: Protein quantitation was carried out by two independent methods:, 1. UV spectroscopy at 280 nm using the absorbency value of 0.47 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). , 2. Analysis by RP-HPLC, using a standard solution of IL-19 as a Reference Standard. Solubility: It is recommended to reconstitute the lyophilized Recombinant IL-19 in sterile 18MOmega-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions. Stability: Lyophilized Mouse Interleukin-19 although stable at room temperature for 3 weeks, should be stored desiccated below -180C. Upon reconstitution Recombinant Mouse IL-19 should be stored at 40C between 2-7 days and for future use below -180C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.
