Product information "Histone H2A.X"
Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene encodes a member of the histone H2A family, and generates two transcripts through the use of the conserved stem-loop termination motif, and the polyA addition motif. Source: Recombinant protein corresponding to human histone H2A.X, expressed in E. coli. Applications: Suitable for use in Enzymatic Assay. Other applications not tested. Recommended DIlution: Enzymatic Assay: Can be used as a substrate for in vitro enzymatic reactions, such as phosphorylation at Ser139 by Rsk1 or MSK1 (M4692-53A). Use 50-100ng /assay. The phosphorylated Histone H2A.X can be recognized by H5110-08C. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile buffer or ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
