A 6kD polypeptide growth factor initially discovered in mouse submaxillary glands. Human epidermal growth factor was originally isolated from urine based on its ability to inhibit gastric secretion and called urogastrone. Epidermal Growth Factor exerts a wide variety of biological effects including the promotion of proliferation and differentiation of mesenchymal and epithelial cells. Recombinant Human Epidermal Growth Factor produced in Pichia is a single, glycosylated polypeptide chain containing 51 amino acids and having a molecular mass of 6kD. , Amino Acid Sequence:, The sequence of the first five N-terminal amino acids was determined and was found to be Asn-Ser-Asp-Ser-Glu, which agrees with the sequence of native human EGF. N-terminal methionine has been completely removed enzymatically. Protein Content: Protein quantitation was carried out by two independent methods: , 1. UV spectroscopy at 280nm using the absorbency value of 2.858 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). , 2. Analysis by RP-HPLC, using a calibrated solution of human EGF as a Reference Standard. Biological Activity: Recombinant Human EGF is fully biologically active when compared to standards. The ED50, calculated by the dose-dependent proliferation of murine BALB/c 3T3 cells (measured by 3H-thymidine uptake) <0.1ng/ml corresponding to a specific activity of 1x10e7 U/mg. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile dH2O, 0.1% HSA or BSA. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
