N-ethylmaleimide-sensitive factor (NSF) is an ~83 kD protein (predicted MW) which was initially isolated based on itís ability to restore intercisternal Golgi transport to N-ethylmaleimide-treated membranes (1). NSF was subsequently discovered to be involved in a variety of membrane fusion events. NSF is an ATPase with two ATP binding domains that are essential for membrane fusion and homo-oligomerization (2). In order to interact with target membranes, NSF requires another set of soluble proteins called Soluble NSF attachment proteins (SNAPs) (3). NSF has been shown to play a prominent role in synaptic vesicle fusion. Together with synaptotagmin and alpha-SNAP, NSF modulates the interaction of SNAP25, VAMP, and syntaxin, in an ATP-dependent manner, to form a complex with a sedimentation coefficient of 20 S (4). This complex may represent an intermediate involved in synaptic vesicle docking and fusion. Applications: Suitable for use in Western Blot and Immunohistochemistry (paraffin). Other applications not tested. Recommended Dilution: Western Blot (Colorimetric) (5,6,7): 0.25ug/ml, Optimal dilutions to be determined by the researcher. Positive Controls: Rat Brain Tissue Extract, Mouse Brain Tissue Extract, HeLa Heat Shocked Cell Lysate, Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20°. Aliquots are stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
