Metallothioneins (MTs) are a class of ubiquitous, low molecular weight, cysteine-rich, metal binding proteins that function in the metabolism of zinc and copper and protects against exposure to toxic metals such as mercury and cadmium as well as a variety of other stress conditions. MTs are present throughout the animal kingdom and are synthesized by nearly every tissue. There are four mammalian MT isoforms: MT-I, -II, -III, -IV and each is encoded by an individual gene. MT-I and MT-II are ubiquitously expressed and differ by a single amino acid charge at neutral pH, whereas MT-III is a brain specific isoform (6) and MT-IV is expressed only in differentiating squamous epithelia. MT expression is induced by heavy metal cations, starvation, elevated temperature, lipopolysaccharide, inflammatory cytokines, free radicals, and ionizing radiation. Several human disorders, such as Indian Childhood Cirrhosis, Menkes' disease, and Wilson's disease, are caused by impaired copper regulation and thus have been linked to altered MT regulation. MT has also been demonstrated to possess immunomodulatory activity. , , Applications: , Suitable for use in ELISA, Immunohistochemistry and Western Blot. Other applications have not been tested. Recommended Dilutions: Immunohistochemistry: Paraffin sections , Western Blot: 1:1000 using ECL. , Optimal dilutions to be determined by researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
