Anti-MSH2 (DNA Mismatch Repair Protein Msh2, hMSH2, MutS Protein Homolog 2)

MSH2 is a 100-106kD member of the mutS family of DNA mismatch repair molecules. It is a DNA binding protein that is expressed in rapidly proliferating cells, and acts in concert with multiple subunits. MSH2 forms a heterodimer with MSH6, forming MutS. This dimer recognizes and repairs both nucleotide misparings and one or two aberrant nucleotide insertions/deletions. MSH2 also heterodimerizes with MSH3, forming MutSB. This complex acts on DNA double-stranded breaks, and repairs large nucleotide insertions/deletions (<15 bases). MutSA is associated with BLM:p53:RAD51 complexes, while MSH2B is associated with SLX4/BTBD12 complexes. Human MSH2 is 934aa in length. It contains three MutS domains aa18-132, 158-284, 297-612 plus an ABC transporter signature motif that may hydrolyze ATP aa 633-852. There are multiple splice variants. One shows a premature truncation after His429, while others contain a 46aa substitution for aa879-934, a 29aa substitution for aa462-934, a 28aa substitution for aa783-934, a 3aa substitution for aa532-934, a 5aa substitution for aa482-934 and a 31aa substitution for aa379-934. Over aa1-140, human MSH2 shares 94% aa identity with mouse MSH2. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 2ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
