Amyloid beta (ABeta) precursor protein (APP) is a 100-140kD transmembrane glycoprotein that exists as several isoforms. The amino acid sequence of APP contains the amyloid domain, which can be released by a two-step proteolytic cleavage. The extracellular deposition and accumulation of the released Abeta fragments form the main components of amyloid plaques in Alzheimer's disease (AD). APP can be phosphorylated at several sites, which may affect the proteolytic processing and secretion of this protein. Phosphorylation at Thr668 (a position corresponding to the APP695 isoform) by cyclin-dependent kinase is cell-cycle dependent and peaks during G2/M phase. APP phosphorylated at Thr668 exists in adult rat brain and correlates with cultured neuronal differentiation. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000. Incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight., Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
