Glutamate is a key molecule in cellular metabolism. It is the most abundant excitatory neurotransmitter in the vertebrate nervous system. Nerve impulses trigger release of glutamate from the pre-synaptic cells. In the opposing post-synaptic cell, glutamate receptors, such as the NMDA receptor, bind glutamate and are activated. Glutamate works not only as a point-to-point transmitter but also as a spill-over synaptic crosstalk between synapses. This Amplite(TM) Fluorimetric Glutamic Acid Assay Kit provides one of the most sensitive methods for quantifying glutamic acid. The kit uses Amplite(TM) Red to quantify the concentration of glutamic acid, which is related to the production of hydrogen peroxide in the glutamate oxidase-mediated enzyme coupling reactions in the presence of glutamic acid. The amount of glutamic acid is proportional to the concentration of hydrogen peroxide formed in the enzyme coupling reaction cycle. In the presence of peroxidase, the fluorescence intensity of Amplite(TM) Red is proportional to the formation of hydrogen peroxide that can be converted to the concentration of glutamic acid. The assay can be readily read with a fluorescence microplate reader at Ex/Em = 540/590 nm. Alternatively the assay can also be read at 570 nm with an absorption microplate reader, but the sensitivity is reduced by about 10 times.
