Helped to reduce background.
Prior to using the Rockland Blocking Buffer for Fluorescent Western Blots, we had been having problems with sensitivity and background. Using conventional milk or BSA blocking left large amounts of background on the LiCOR imaged PVDF membrane. Using the Rockland Blocking Buffer for Fluorescent Western Blots prior to adding primary, we were able to drastically reduce the amount of background in order to see CFTR in Calu-3 cells in addition to other cell types where CFTR protein amount is greatly decreased. I would recommend not adding tween to the blocking media and to make sure to have ample washing steps.
