The normal crossreacting antigen (NCA) or CEACAM6 belongs to the carcinoembryonic antigen (CEA) gene family. CEACAM6 is a GPI-anchored membrane protein with a single N domain, which shows structural homology to the immunoglobulin variable domains, followed by two immunoglobulin constant-like domains.

Flow Cytometry: 1:50-1:100
Optimal working dilutions to be determined by researcher.
Flow Cytometry Use 10ul of the suggested working dilution to label 10e6 cells or cells or 100ul whole blood.
Immunohistochemistry: Requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose. Recommended positive control tissue: Colonic cancer.
Recommended Secondary Reagents F(ab')2 rabbit anti-mouse IgG HRP, F(ab')2 rabbit anti mouse IgG: RPE conjugate
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

SpecificityRecognizes human CD66c and CD66e, members of the carcinoembryonic antigen family (NCA and CEA respectively. In flow cytometry this antibody stains peripheral blood granulocytes, but not monocytes or lymphocytes. In IHC the antibody stains a range of tumor cells and also neutrophils and some endothelial cells. Cross-reactivity with other members of the CEA family has not been reported, but has not been formally excluded.
