Caspases: New Roles for Necroptosis & Immunity Programmed Pathways Beyond their well-studied role as mediators of apoptotic programs, caspase participation in regulation of additional cellular pathways is now being characterized in a variety of far-ranging cellular processes. In regulation of innate immunity, caspase-8 has been implicated in regulation of pro-inflammatory cytokines, in adaptive immunity in T-cell proliferation, and in preventing NECROPTOSIS, an emerging necrotic cell death pathway, among other activities1-2. Caspase-mediated induction of apoptosis may in fact also lead to compensatory proliferation3. Caspases are also being studied in cell differentiation mechanisms related to a number of cells including stem cells, where caspase-3 cleavage of Nanog, a master transcription factor needed for self-renewal, may lead instead to differentiation4. Caspase-11, especially, and caspase-1 appear to function in regulating cell migration and motility through actin formation and polymerization or actin dynamics5-6.
