Sekundärantikörper

Sekundärantikörper ermöglichen den indirekten Nachweis eines Antigens, indem sie an der konstanten Region des Primärantikörpers binden. Voraussetzung hierfür ist, dass der Sekundärantikörper eine Reaktivität gegenüber der Spezies (bspw. Maus, Kaninchen oder Ziege) und der Klasse  (bspw. IgG oder IgM) des Primärantikörpers aufweist. Für das Labeling von Sekundärantikörpern werden häufig Enzyme (bspw. alkalische Phosphatasen) gewählt. Diese setzen Substrate zu farbigen Produkten um und dieser Prozess kann photometrisch ausgewertet werden. Neben der Verwendung von Enzymen ist die Fluoreszenzmarkierung von Antikörpern mit Farbstoffen, wie etwa mit FITC oder DyLight 488, eine verbreitete Labeling-Methode.
