Product information "Anti-acetyl-Histone H4 (Lys12)"
Applications: , Suitable for use in Peptide Inhibition, Peptide Dot Blot, Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Peptide Inhibition: 10uM of a histone H4 peptide containing acetyl-lysine 12 abolished detection of histone H4 by anti-acetyl-Histone H4 (Lys12) in immunoblot analysis of HeLa acid extracts. Little to no signal reduction was observed with 10uM of histone H4 peptides containing acetyl-lysine 5, acetyl-lysine 8 or acetyl-lysine 16. Peptide Dot Blot: A 1:1,000 detects serial dilutions (25, 5, 1 and 0.2uM) of a histone H4 peptide containing acetyl-lysine 12 spotted onto PVDF, and not histone H4 peptides containing acetyl-lysine 5, acetyl-lysine 8 or acetyl-lysine 16. , Western Blot: 1:3,000 to 1:10,000 detects acetyl-Histone H4 (Lys12) in acid extracted proteins from HeLa cells. The antibody did not detect unacetylated recombinant Histone H4. Immunoprecipitation: 2-6ul immunoprecipitated the Ime2 promoter from wild type yeast chromatin and not from mutant yeast chromatin containing a histone H4 K12R mutation. , Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
