Anti-PA28, gamma, NT (Proteasome Activator 28-gamma subunit)

Proteasome activator 28 Supplied as a liquid in PBS, pH 7.4, 0.1% sodium azide.(PA28), also known as 11S regulator, is composed of two homologous subunits, alpha (a) and beta (b), that share ~ 50% amino acid identity. A separate but related protein, PA28gamma, was originally identified as Ki antigen from sera of patients with systemic lupus erythematosus. PA28a and PA28b are located primarily in the cytoplasm, whereas PA28g is located in the nucleus.4,5 PA28 binds to the outer rings on both ends of the 20S proteasome to form a football-like structure.6,7 Binding of PA28 greatly stimulates multiple peptidase activities of the 20S proteasome in an ATP-independent reaction, but lacks the ability to degrade large protein substrates, suggesting that PA28 may cooperate with the 26 S proteasome in a sequential proteolytic pathway.8,9 Abnormally high expression of PA28g has been observed in thyroid cancer, particularly in regions of cancer cells where growth rate is accelerated. PA28g interacts directly with Hepatitis C virus (HCV) core protein, which becomes retained in the cell's nucleus,12 and with MEKK3, which phosphorylates PA28g and increases its cellular levels. Applications: Suitable for use in Western Blot, Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1-3ug/ml., Immunoprecipitation: 7ug/reaction, Optimal dilutions to be determined by the researcher. Positive Control: human MeWo malignant melanoma, A375 malignant melanoma, Jurkat T-lymphocyte. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
